Expression of the novel tumour suppressor sterile alpha motif and HD domain?containing protein 1 is an independent adverse prognostic factor in classical Hodgkin lymphoma

The expression patterns and prognostic significance of sterile alpha motif HD domain-containing protein 1 (SAMHD1) in the neoplastic Hodgkin Reed Sternberg (HRS) cells lymphoma (HL) were investigated a cohort 154 patients with HL treated standard regimens. SAMHD1 was assessed by immunohistochemistry using diagnostic lymph node biopsies obtained prior to treatment. Using an arbitrary 20% cut-off, positive HRS 48/154 (31·2%) patients. not associated clinicopathologic parameters, such as age, gender, stage or histologic subtype. In 125 median follow-up 90 months (7–401 months), significantly correlated inferior freedom from progression (FFP) (P = 0·025), disease-specific survival (DSS) 0·013) overall (OS) 0·01). Importantly, multivariate models together disease stage, histology subtype type treatment covariates, retained independent significant association unfavourable FFP 0·005) well DSS 0·022) OS 0·018). These findings uncover novel, adverse factor that may have therapeutic implications since inhibitors are now available for clinical use.
